B. Hossain, A. Y. Mohammad and A. I. Ali College of Veterinary Medicine\ University of Anbar Summary The study was conducted on 36 local rabbits divided in to two groups, the first group was subdivided in to three groups, the first and second groups of them injected with body extract of T. canis intradermally (0.ml and 0. ml respectively) while the third group injected with phosphate buffer saline and considered as control group. The second group also subdivided in to three groups, the first and second groups injected with same antigen intraperitonealy and intramuscularly in dose of 0.4 ml and 0.3ml respectively, after 4 days they reinjected by boosters dose, while the third group leaved untreated and considered as control group. Our results showed a significant difference in hypersensitivity test of immunized groups with an average of skin thickness has reached to 3.8 and 4.7 mm after 4 hours and an average of redness diameters 3.84 and 4.4mm after 4 hours in comparisons with the control group.36 and 0 mm. Our finding showed valid decrease in weight of liver and spleen in immunized groups and reach an average of (.9 and 0.09 gm) and (.89 and 0.08 gm) respectively, in comparisons with control group (3.50 and 0.04 gm). There was a significant increase of IgA and IgG levels in immunized groups and reach an average of ( and mg/dl) and ( and mg/dl) respectively, in comparisons with the control group (9.66 and mg/dl). There was valid decrease of neutrophils numbers of immunized groups, while there was a significant increase in lymphocyte numbers of immunized group in comparisons with the control group. It was concluded from this study that immunization against body extract of Toxocara canis could induce immune response in local rabbits. 75

Yaseen*** * College of Veterinary Medicine\ University of Anbar ** College of Veterinary Medicine\ University of Baghdad *** Iraqi center for cancer and medical genetics research\ AlMustansiriyah University Summary This study is considered the first in the country on growing of bovine papilloma in cell culture from skin warts of cattle in order to detect papilloma virus, and to establish a cell line of transformed cells for further studies. In this study the warts were surgically removed from cows showing lump lesions on skin of abdomen, neck and udder, and transferred aseptically to laboratory by transport media. Trypsin and collagenase enzyme were used to dispersed papilloma cells from fragments of warts, Dulbecco s modified Eagle s medium (DMEM) and Rosswell Park Memorial Institute (RPMI)640 Medium (Gibco, were used with 0% fetal calf serum for culturing of papilloma cells. Cultured cell were first noticed after 34 days post incubation(pi) and appeared as epithelial and fibroblastic cells. After 7days post inoculation clones of transformed epithelial cells start to appear and after (05) days it became a complete monolayer. Successful secondary culture was achieved by using trypsinversene solution(tv). Bovine papillomavirus (BPV)associated diseases are important in veterinary medicine and can also be considered as possible important models for the study of human papillomavirus (HPV). Often, immunosuppression in cattle results from exposure to bracken fern: some animals become unable to reject the infection and succumb to widespread cutaneous or mucosal involvement (). These stages of papillomatosis are problematic leading to economic losses (3, 4). Six types of BPV have been described (6) (5), and recently, four other types were reported. BPV causes teat and penile fibropapillomas; BPV is associated with cutaneous warts, alimentary fibropapillomas and urinary bladder tumors; BPV3 causes cutaneous papillomas; BPV 4 is associated with pure epithelial papillomas of the upper gastrointestinal tract; BPV5 induces fibropapillomas of the udder; BPV6 causes papillomas of the teats; BPV8 causes cutaneous papillomas; BPV9/0 are associated with epithelial squamous papillomas of the udder (). BPV is described as epitheliumspecific (6). However, the presence of viral DNA sequences in different cells such as those of peripheral blood and gametes has been reported (7). The papilloma virus Belong to family papillomaviridae produce in their hosts benign skin tumors (papillomas);which contain variable amount of infectious virus (). Different methods have been used to treat bovine papillomas. A formalinized suspension of bovine warts with inactivated virus provides a vaccine for effective treatment and prophylaxis of bovine papillomatosis. In Iraq, AlBana and Khazeil (8) prepared the bovine interferon and used it for treatment of bovine papilloma for the first time in country. Our aim of this study was to establish a cell culture system for the bovine papilloma in order to specifically identify the virus and to prepare a vaccine from the cultured papilloma cells. Materials and Methods Collection of wart samples: Cauliflower like wart were collected from cow suffering from papilloma skin lesion on the udder, teat, abdomen and the back of affected animals. Before surgical removal of the warts the animal were given ml of xylazine % (0.50.5ml/00kg.b.w), as relaxant intramuscularly and lidocaine % as local anesthesia infiltrated around the lesion, followed by cleaning with normal saline. Aseptically the warts were removed by surgical procedure. Pieces of the lesion were added to transport solution containing MEM medium supplemented with a mixture of Antibiotics (amikasin, streptomycin and amphotricin) in sterile containers. Other small pieces were added to 0% formalin in plastic containers for histopathological examination. Dispersion of papilloma cells: The papilloma pieces were first washed thoroughly with sterile phosphate saline (ph 7.) and MEM medium to remove the dirt and extraneous material covering the lesions. The papilloma were cut into small pieces by sterile scissors and washed again then divided into two parts: The first part was added to trypsin 0.5% solution and second part was added to collagenase enzyme 0 mg/ml, was added (00 μl),at 37 C. Trypsin part was allowed to disperse the papilloma cell for 30 minutes by continuous stirring, while, collaginase part was allowed for 40 minutes by continuous shaking. The dispersed cells in both methods were filtered by using sterile gauze followed by centrifugation at 000 rpm for 5 78

The sedimented cells were mixed with two kinds of media RPMI, MEM supplemented with 0% fetal calf serum, then cells were seeded in a disposable plastic flask (5 cm ) and incubated at 37 C in Co incubator. Subculture of the Primary Papilloma Cells: TrypsinVersene Solution (US Biological, USA) was prepared by dissolving 0g of trypsinversene (TV) powder (USbiological, USA), in one liter PBS, then sterilized by Nalgen filter (0.μm). The primary monolayer papilloma cell culture were dispersed from the surface of the cell culture flask by using TV solution (at 37 C) and then seeded into new flask after mixing the cells with growth MEM medium and incubated at 37 C in CO incubator. Results and Discussion Growth of cultured of papilloma cells: Trypsin dispersed papilloma cells attached to the bottom surface of cultured flasks were first noticed after 487 hr. post incubation (PI) at 37 C. These cells started to multiply after 4 days (PI) and appeared as epithelial and fibroblastic cells. After seven days (PI) clones of transformed epithelial cells started to appear with new fibro plastic cells as the periphery of these clones (Fig. ). Other part of the cultured flask showed fibroblastic cells only. The clones of transformed cells becoming larger in size with multi layer cells center and these fibroblastic cells at the periphery focus (Fig., A and B). Our results are in agreements with (6) which indicate the culturing of fibroblastic cells only from papilloma lesion without presence of clones of transformed epithelial cells (Fig. 3). After 05 days post culturing of papilloma cells the whole surface area of flask were filled with cultured cells and look like multilayer of transformed cells (Fig. 4). Results of using collagenase enzyme for dispersion of papilloma lesion showed fibroblastic type of cells only started at 79 days post incubation without epithelial cells (Fig. 5). After confluent cell cultured reached papilloma cells were sub cultured by using (TV) solution and successfully a second passage of the cells result in duplicate culture flasks and after 3 days both type of epithelial clone cells and fibroblastic cells were noticed. Kh. Jalob, M. D. Taha, M. M. Jumaa, D. A. Najim & A. A. Salman College of Veterinary Medicine\ University of Anbar Summary The study aimed to evaluate the immune response for NDV depending upon clinical cases brought by the client to the clinic of college of veterinary medicine, University of Anbar, during the period from October, to December 3,00. The study was conducted on 86 broiler chicks. The animal were divided into 3 groups according to their age. Haemagglutination inhibition (H.I) test was the criteria of evaluation the immune response. Blood samples were collected via direct slaughtering of chicks. Serum samples were separated for immunological test. Group : Included 5 sample, the age ranged between 47 days, they had got one time vaccine. Group : Included 44 sample, the age ranged from 0 0 days, they have got two time vaccines. Group 3: Included 7 sample, the age ranged from 030 days, they have got three time vaccines. The result showed that low Abs titer have been detected in group which showed a marked relapse in their immune status as expressed by the mean value ±87.64 of H.I titer. There were a significant difference between the third group and other groups (P<0.05). Sci., Vol.: 4, Supplement, 0 ISSN: Proceedings of First Medical Conference of Medical Colleges (Veterinary Research)\ University of Anbar Introduction Newcastle disease virus (NDV) which is an important viral disease is the main health barrier for international trade of poultry and poultry products (). Now a day ND has a worldwide distribution and affects several birds species (). The virulence of ND strains varies greatly with the host (3), but breed or genetic stock does not appear to have a significant affection on the susceptibility of chickens to the disease (4). Therefore mass application of vaccines in addition to biosafety measures are needed for its control and vaccination practice became a routine work throughout the world (5). In our country the most common program adapted is through exposure of chicks to three times of vaccines in different route till the time of slaughter. Such type of vaccination program showed a failure from time to time creating sever economic losses making the possibility for this disease to threat the process of poultry industry, for this reason our study have been directed to look for this point (vaccine failure) through evaluation the immune status brought to the clinic by means of H.I test and to build a scientific base for our client to give a recommendation about the employed vaccination program servicing our national poultry industry. Materials and Methods Animals: A total of 86 broiler chicks brought to clinic of college of veterinary medicine\ University of Anbar by the clients for diagnostic purposes. The samples were received from 3 poultry houses distributed randomly in AlAnbar province. Chicks were subdivided into three groups: Group: Included 5 birds at the age from 40 days, they have received one time vaccine. Group : Included 44 birds, the age ranged from 00 days, they have got two times vaccine. Group 3: Included 7 birds, the age ranged from 030 days, they have received three times vaccine. Serum: Blood was taken through direct slaughter in a special test tube with an average of 5 ml from each birds. The sample were left at 4 ºC in the refrigerator from 54hr. The serum was separated according to Hanson, R.P (6). Washed RBC: Hepranized tubes were used for collection of blood from slaughtered chicks, ml of blood was collected in each test tube to which 8 ml of 0.9% Nacl solution was added, the mixture was centrifuged for (0) minutes at a speed of 500 r.p.m. The process was repeated three times and % of the suspended RBC was used for Haemagglutination inhibition test. Haemagglutination (HA) test: Materials used in this technique included: Saline solution % suspension of washed RBC, Perspex agglutinating tube, ( ml) disposable syringe. Two fold dilutions was made for the seed virus. The potency of virus was evaluated according to Hanson, (6) for further use in haemagglutiation inhibition (H.I) test. Titration of the virus: One vail of Newcastle disease vaccine Lasota strain was taken from a private clinic and was titrated according to Reed and Muench (7). The result was identical to that fixed in the label (i.e) EID Haemagglutination inhibition (HI) test: Beta method was the technique followed through which the serum have got two fold dilution while the amount of diluted virus was fixed. The mixtures were left 3040 minutes until the control well showed a settled RBC. The end point was indicated by the highest dilution giving inhibition (8). Statistical analysis: It was done according to SAS (9) and Duncan(0). A significant level of 5% was used. 83

Sci., Vol.: 4, Supplement, 0 ISSN: Proceedings of First Medical Conference of Medical Colleges (Veterinary Research)\ University of Anbar Result and Discussion A total of 86 serum samples were collected from different commercial broiler farms and were subjected to H.I test. Group : Showed a variable level of Ab titer ranging from 0 to with a mean value of ± (Table ), such high range of variability was related to the different sources of their parent stock which would be the source of further differences within the same individuals of other groups, also this gave an explanation for high value of standard error (SE) in this group. The mean value of group indicated that such newly hatched birds posses high level of M.I which is protective against infection for the first few 4 days. This result agreed with Jalil etal () Who regarded that a titer of 7 is protective against virulent challenge while Allan and Gough (8) explained that a titer of 3 is generally accepted as positive for specific immunity against ND. This point is the major problem for the vaccine failure because of their neutralizing effect with the vaccine Ag rendering the vaccine ineffective (,3,4,5). The effect of maternally derived antibodies (MDA) in the interference with the replication of vaccinal particles lead some companies to shift their vaccination strategies from the preferred 74 days of age by drinking water in broiler flock to coarsespray at day in the hatchery (6), this is because by such way maternal antibodies did not influence the development of immunity (7). Group : Showed the most critical period in the immune status of the birds in which the HI level in the collected samples had a variable range from 0 (Table ). Samples showed HI titer of 0 (not protective) were 3.3%, 3.6% and 7.4% in group, and 3 respectively. On the other hand the mean value in group was the lowest among other groups ± (Table 4). Similar findings had been observed by Numan et al (8) who explained that the causes of relapse in such cases might be due to several reason such as poor vaccine quality, unsuitable vaccination schedules or vaccination techniques, impaired immunecompetence due to immunosuppressive substance in the feed or to immunosuppressive disease and therefore unable to protect the chick from NDV infection. We believe that the vaccine failure in this group was related mainly to the high level of M.I that interfere with the process of vaccination by neutralizing the vaccinal particles specially the routine work through drinking water (D.W) (9). The intensive vaccination program is also regarded as a main cause for vaccine failure specially with those birds possessing high passive immunity. It is well known that newly hatched chicks with HI level of 7 should not be vaccinated before 8 days old, since the HI level decrease with an average of Log\ 4.5 days (0,). Another important point for explanation of vaccine failure is the type of vaccinal strain. It is well known that AG68L strain have been proven to be of considerable potential as lentogenic vaccine for administration in the D.W as reported by Kaschula (). The most important character of this strain in that it does not interfere with MDA by which the vaccinator could overcome the effect of M.I (3). Group 3: Showed a significant difference in the mean value ± 57.9 of HI titer when compared with other groups (Table 4). The number of samples collected which had Hi titer above 6 (Table 4) were samples giving an indication of protective level 8.4%. This result agreed with Jalil etal () who mentioned that HI MDA titer of 8 gave protection to chickens while birds below 8 could not with stand the challenge infection with virulent NDV. Also our results correspond to that mentioned by Box (4) who suggested that HI titer should be 7 log (i.e:8) or more for protection against ND, on the other hand the protective level in group and were 46.6% and 45.5% and both level were unsatisfactory. An important feature of group 3 as shown by (Table 3) is their wide range of 84

In conclusion, there is no fixed vaccination program, it depend upon the level of MDA. In endemic area the effect of havoc to ND on poultry industry could be minimized by coarsespray in one day old and increase the gap period between this vaccine and the booster dose for a period not less than 4 days. AlBadrani College of Veterinary Medicine\ University of Mosul Summary This evaluation of some hematological and biochemical changes associated with chronic toxicity of Nerium oleander aqueous leaf extract twenty rabbits of both sexes were made. First group treated orally with Nerium oleander aqueous leaf extract at a dose rate of 0 mg\ kg body weight daily for 4 months, while animals of the second group do not left as control. The main clinical signs observed as anorexia, nervous signs, restlessness, crying, ataxia, pawing of the ground, convulsion, falling, turning of the head back ward, polyuria, emaciation, increased heart sound intensity, noisy respiration, paralysis associated hind limbs extension, finally death. Hematological changes increased in the packed cell volume (Hemoconcentration), and hemoglobin concentration, and erythrocytic count and leukocytosis with lymphocytosis, eosinophilia, and neutropenia were noted. It is an ornamental shrub, densely branched tree, to 0 m tall. (). This plant grows outdoors in warmer regions, and sometime it is grown as a house plant. It is widely cultivated in Mosul (Iraq) along roadsides, edges of woods and gardens. This extremely 88

The toxic principles are two potent cardiac glycosides, oleandrin and neriine (Cardenolides), and can be isolated from all parts of the plants (). Common oleander contains a strychinine like toxin, and a heartactive cardiac glycosides substance (similar to the digoxin) (3, 4). Apparently the plant is not palatable, but will be eaten by hungery animals (). There are records that the plant can be used as a rodenticide, insecticide and for indigestion, fever, ringworm, leprosy, venereal diseases (5), also as cardiac drugs (3, 6), and antidiabetic agent (7). Livestock are usually poisoned when they are allowed to graze in places where oleander is abundant or when pruning are carelessly throun into animal pens (8). Seven outbreaks of acute intoxication from oleander in cattle were reported in Northeast of Brazil (9). The minimum lethal dose of oleander for cattle is 50 mg\ Kg body weight (0). Horse given 40 mg\ kg body weight of green leaves via nosogastric tube consistently developed severe gastrointestinal and cardiac signs of poisoning (). Single oral doses of or 0.5 g of dried N. oleander caused restlessness, chewing movements of jaws, dyspnea, ruminal bloat incoordination of movements, limb paresis and recumbancy and death 4 4 hr. after dosing, while the daily oral doses of 0.06g (60 mg\ kg) dried N. oleander leaves\ kg body weight caused less severe signs and death occurred between day 3 and 4 (). Asigle oral lethal dose of 0 mg of dried N. oleander leaves\ kg body weight began to appeared the clinical signs of toxicosis in sheep about 30 minutes after exposure and animal died within 4 4 hr. (3). Multiple exposure of the mice to the dose 000 mg\ kg of 70 % ethanol extract of the N. oleander dry leaves was injected subcutaneously once a week for 9 weeks failed to express a significant influence on blood parameters as well as myocardium. On other hand a lethal dose (4000 mg\ kg body weight) was capable of inducing progressive changes in myocardial electrical activity ending up in cardiac arrest (4). One of 0 rabbits in an experimental study inducing chronic cardiomyopathy after treated with cardiac glycosides at 3 mg\ kg body weight intravenously in the lateral ear vein once a week for 6 weeks period (5). The median lethal dose in rabbits subcutaneously injected with N. oleander aqueous leaf extract was mg\ kg body weight (6). From a review of the literature it become clear that chronic toxicity with N. oleander aqueous leaf extract has not been described. Therefore, the purpose of this study was to record the hematological changes and their correlation with biochemical changes in the chronic toxicity of Nerium oleander aqueous leaf extract in rabbits. Material and Methods Animals: The study conducted on the twenty local breed rabbits, of both sexs, year age,.5 kg. Preparation of plant extract: Nerium oleander fresh green leaves were used. Leaves were collected from plants growing in the in Mosul city (Iraq) from different localities on roadsides and gardens during May and June. The plant was properly identified. Fresh plant leaves were washed with distilled water. A 500 g quantity of the plant material was cut into small pieces and blended using an electrical blender with 500 ml of 0 m M potassium phosphate buffer (ph 7.). The mixture obtained was pressed through acheesecloth and the filtrate was centrifuged at 0000 xg for hour. The supernatant fluid was separated and sterilized by filtration through nitrocellulose membrane (pore size 0. µm) obtaining a clear solution, which was dried by lyophilization. Sterile extract were stored at 0º C until used (7). The study methods: Animals were divided into two groups of 0 rabbits each. Animals of the first group were treated orally with of Nerium oleander aqueous leaf 89

The extract was dissolved in 5 ml of phosphate buffered saline (PBS). The dose of the Nerium oleander extract was based on previous toxicological studies (5, 6). Animals of the second group (control groups) were treated with equal volume of PBS daily for 4 months. All animals kept under daily observation and their hematological and biochemical changes were examined at monthly intervals. Samples:. Whole blood sample with anticoagulant (disodium salts of EDTA): About ml of whole blood samples were collected from each animals of both groups in dry clean tubes and were used for measurement of the packed cell volume (PCV), heamoglobin concentration (Hb), total red and white cell counts (TEC, TLC) and differential cell count (DLC) by using coulter counter and according to (8).. Blood serum samples: Blood serum samples were taken from heart in a dry, clean and sterile centrifuge tubes. The samples were allowed to be clotted at room temperature. The clotted blood were centrifuged at 3000 rpm for 0 minutes. A clear sera were separated by Pasteur pipette and tranfered into a clean, dry and sterile stoppered glass vials till peforming the biochemical analysis. Determintion of total serum protein, albumin, globulin, albumin globulin ratio, blood urea nitrogen, creatinine were done by using commercial standard Kits (BioMerieux, Baines, France) (9). Statistics: The statistical analysis were determined using Student, s t test. A P value less than 0.05 was taken as significant (0). Results The clinical signs of toxicosis in the rabbits began to appear in 30 th days after the exposure to the extract included anorexia anorxia, nervous signs, restlessness, crying, ataxia, pawing of the ground, convulsion, falling, turning of the head back ward, polyuria, emaciation, increased heart sound intensity, noisy respiration, and five rabbits appeared paralysis associated hind limbs extension and died within 3 days in 90 th days of the study, while other treated animal died between days 00 and 0 of the study. The hematological changes in treated rabbits began to appear in 30 th days and persist to 90 th days after the exposure to the extract included a significant increase in packed cell volume (heamoconcentration), hemoglobin concentration values and total erythrocytes and leukocytic counts. The highest value was reached on 60 th day post treatment (Table ). The effect of the extract on the differential leukocyte counts included, a transient increase in the lymphocytes number (lymphocytosis) was registered together with (neutropenia). Eosinophils significantly increased on 30 th onward. No changes were observed in the numbers of either monocytes or basophils (Table ). The results of the biochemicals tests revealed to significant increase in the total protein and globulin concentration in serum of the treated rabbits from day 60 th onward. The highest rate recorded in the 90 th days of the study. Also showed decreased in albumin in the serum of the treated rabbits from day 60 th onward, till the end of the study the albumin\ globulin ratio also decreased from day 30 th of the study (Table 3). The results also showed a significant increased in the blood urea nitrogen and creatinine levels from day 30 th onwards in the animals of the first group, and till the ends of the study as compared to the values of the treated animals in the day 0 (pre exposure) and animals of the control group (Table 3). Rabbits was an animal model of low output cardiac failure with activation of vasoconstrictor mechanism (). The common clinical signs of oleander toxicosis in cattle included locomotion disturbances, diarrhea, depression, and sudden death (9) while in sheep included dyspnea, grunting, salivation, grinding of teeth, ruminal bloat, frequent urination, ataxia and recumbancy prior to death (). Hemoconcentration and leukocytosis began to appear after 30 days of the study and persist to the 90 th days of the study this is may have been associated with shock and dehydration (3). 9

Lymphocytosis and eosinophilia with neutropenia also recorded in the our study. Lymphocytes play an important role in the immune response and their number in circulation is increased during chronic infections or chronic toxicity. However, neutropenia along with eosinophilia are generally observed in chronic inflammatory conditions (8) Heamoconcentration resulting from these various alterations may mask the existence of anemia and interfere with proper interpretation of both total erythrocyte and leukocyte counts (8, 4). Hyperproteinemia, hypoalbuminemia and a significant increased in globulin concentration in serum were also recorded in the our study. The hyperproteinemia usually recorded in the dehydrated animals, and in animals were suffered from anorexia and their livers were not efficiently synthesizing protein, the elevated globulins substituted the reduced albumins and thus total protein values were observed usually with liver diseases. Hypoalbuminemia may be altributed to inhibition of its synthesis, its rapid breakdown and its losses (8, 3, 4). The results also showed gradually significant increased in the blood urea nitrogen and creatinine levels as result of renal impairment. The damage of kidneys facilitate the retention of blood urea nitrogen and creatinine (8, 5). The main lesions in sheep treated with daily oral doses of 0.06 g dried. N.oleander leaves\ kg body weight included hepatonephropathy and gelatinization of the renal pelvis and mesentry and were accompanied by a significant increases in serum AST and LDH activities, bilirubin, cholesterol and urea concentration and a significant decreased in total protein and albumin levels (, ). M. T. AlJumaily** *College of Veterinary Medicine\ University of Anbar **College of Agriculture\ University of Anbar Summary Epididymus were collected from AlRamadi Slaughter house during the periods from September 00 to march 003. The samples were transported to the Laboratory with normal saline at a temp of 3335c within 30 minutes. Semen were collected from the tail of epididymus by aspiration with 8 gauge needle on 3ml disposable suring containing ml TALP media. The collected media together with spermatozoa were put in a Petri dish and incubated in Co incubator at 35 c, 90% Humidity for 46 h. Spermatozoa evaluated for their maturation (The distal Protoplasmic droplet). Matured Spermatozoa were washed with buffer Solution. Three times with Centrifugation. Then 0.5 ml of spermatozoa pluse 9.5 ml of TALP media with 0 mg/ml heparin, Centrifuged, and then 0.5 ml from the bottom spermatozoa incubated in Co incubator at 35c with 45 angle. Head to Head agglutination of Spermatozoa was considered the Criteriia of sperm Capacitation. The results showed that the procedures give a best result for sperm maturation and capacitation in vitro. Capacitation defined as a series of biochemical and biophysical changes prior to fertilization (). Capacitation involves the removal of sperm coating material acquired during epididymal transit or during exposure to seminal plasma and cholesterol depletion resulting in increased membrane permeability to calcium (). In vitro washing of spermatozoa before incubation was reported to accelerate capacitation. Heparin is used for in vitro capacitation of spermatozoa. It modulates gamete interactions in cattle and sheep IVE system (3). The aim of this study was designed to show the capcitation of arm spermatozoa obtained from the tail of epididymis. Materials and Methods Epididymus were collected from AlRamadi Abattoir during the periods from September 00 to march 003. The samples were transported to lab with normal saline with temp 3335c within 30 min, semen were collected from the tail of epididymus by aspiration with 3 ml disposable suring Containing ml of TALP media according to the size of the tail of epididymus according to Im, etal (4). Then the tail of epididymis minced with medical scalpel in order to carryout the sperm in the media. The collected media together with the sperms were pult in a sterile petridish and then transported to be incubated at 35 c. Samples were taken from the media to observe the degree of maturation and evaluate the sperm according to the presence of protoplasm droplet and their position on the sperm. Evaluation of semen included individual motility and sperm abnormalities. The sperms then were incubated at 5% Co,35 c, 90% relative humidity for 46 hours. In a sterile test tubes with 45 angle without cover. In vitro Capacitation of Sperms: Following in vitro maturation and examined under microscope to see the degree of sperm maturation, Then the sperms leaved in the media outside the incubator at room temp (36c) for 30 minutes. After that the sperms were washed with buffer solution (at temp 35c) [nacl; 9 mg/ ml, BSA; mg/ ml, FCS; 0%, Cryst. Penicillin; 00 I.U./ ML, Streptomycin. Sulphate; 0. mg/ml]. ml from semen diluted with washed solution to 0 ml. in a test tubes, then centrifuged at room temp 00 R. per. minte/ for 3 minutes for three successive times. Discard the supernatant fluid and the sperms stays at the bottoms of the test tubes with a little amounts of media. Addition of 0 ml of TALP media [NACL; 4.0mm, KCL; 3.mm, Nahco3; 5.0mm, NaH Po 4 ; 0.3mm, Sodium Lactate;.0mm, Cacl ;.0mm, Mgcl ; 0.5mm, Sodium Pyruvate;.0 mm, Heparin; 0 mg/ml, BSA; mg /ml, Streptomycin Sulphate; 50 mg/ml, Crystalline Penicillin; 00 i.u./ml]. Then Centrifuged again at the same previous manner for one time. 9 ml of supernatant of the media was removed, Incubated in Co incubator for 5 minutes at 35c with 45 angle Head to head agglutination of spermatozoa is considered to be a Criteria of sperm capacitation(). Results and Discussion The results of this study showed the possibility of sperms maturation taken from the tail of epididymis in TALP media in vitro. The presence of distal protoplasmic droplet was the criteria of sperm maturation. Similar observations have been made by Wani (5), Wani, etal. (6) and AlJumaily (7). Several workers indicated that epididymal spermatozoa have been successfully used from slaughtered rams, many hours after their death, for IVF of sheep oocytes (, 5, 7). TALP cultured media for in vitro maturation of sperms were easily prepared media in the Lab (8). This might be attributed to their 95

The result also showed the criteria of head to head agglutination, which indicates the sperm capacitation. It has been observed that heparin stimulates the fertilization rate by improving the efficiency of sperm capacitation in sheep (9). Heparin apparently binds to sperm and plays a role in the sperm uptake of calcium (0). Capacitation involves the alternation of the plasma membrane, such as removal of decapicitation factors, removal of cholesterol, influx of calcium, an increase in intracellular PH, and increase in protein tyrosine phosphorylation. The Majority of changes that occur during capacitation involve sperm head; however no morphological occur. Capacitation is believed to allow the sperm to undergo the a croseme reaction. The life span of capacitation sperm is limited; as a result, precise timing between capacitation and the acrosome reaction is necessary for proper fertilization to occur (3). References. Wani, N. A. Sh. Abid*, M. S. Shallal** and L. H. Ali* *College of Education Pure Science\ University of Anbar **College of Veterinary Medicine\ University of Anbar Abstract This study was conducted to reveal the effects of vitamin E and cholesterol on white New Zealand rabbits. Cholesterol and vitamin E were added in their diets with a certain percent to each group. Other animals were divided into four groups in order to study the resulted histological and biochemical changes. The first group was given a normal diet (no addition of cholesterol or vitamin E), the second and the third groups were given % cholesterol and % cholesterol with % vitamin E, respectively. The fourth group was given % cholesterol for eight weeks till they had diagnosed to have atherosclerosis and then given % vitamin E for four weeks after infection ANOVA table was used to compare the mathematical means through the least significant values test, (L.S.D) among treatments that were replicated three times, at α levels 0.0 and Results showed the followings: The addition of % vitamin E with % of Cholesterol to the diet decreased the average of the total cholesterol (TC), low dense lipoproteins (LDL) and triglycerides (TG). TC values in the first group were 49.8±.93, 53.4±.673, 54.8±.93 and 58.6±.70 mg/dl for the first day of the first week, and the 4 th, 8 th, and th weeks, respectively. TC averages in the second group were 49.4±.408, 353.±3.49, 39.0± and 5.0±4.58 mg/dl., in the third group were 46.0±3.608, 5.0±.449, 89.6±.408 and 76.±3.70 mg/dl.,and in the fourth group were 50.6±.509, 8.0±.449, 40.6±3.847 and.4±4.59 mg/dl., for the first day of the first week, and the 4 th, 8 th and th weeks, respectively. Low dense (LDL) and the very low dense lipoproteins (VLDL) gave similar reactions to the total cholesterol percentages as to their increase in the second group and gave a gradual decrease in the third and fourth groups. High dense lipoproteins (HDL) gave similar averages as to control at the same various specified times of the experiment. The number of foam cells had increased gradually from the fourth week and also, the collagen and elastic fibers in addition to smooth muscles of the endothelium, the reason to which its thickness had increased. The medium tunica had a number of foam cells and other gradual difference in its compact components especially the elastic and other muscular fibers the reasons to which its thickness had decreased.

There was a clear increase in the endothelium thickness, but the thickness in the medium tunica had decreased as compared to control and to the group affected with hypercholesterolemia. AlHassany*** *College of Veterinary Medicine\ University of Baghdad ** AlHilla educational hospital *** Veterinary hospital /Babylon Summery This study was aimed to investigate the prevalence of brucellosis disease in humans and sheep in Al Hashymia district\ Babylon province depending upon data which was register in Babylon hospital and veterinary hospital in Babylon. All suspected cases infected with brucellosis were examined by rose bungal test during The results revealed that the number of positive cases which were entered and register in the hospital were 4 Case, the positive cases of 996 were the highest reached 30 case (53.33%) while the positive cases of 995 were the lowest 9 (5.3%) Also results revealed that Al Madhatia and Ghazali showed the highest positive cases 6 (49.5%) while lowest positive cases appeared in AlTalyha 4 (3.8%). The villages and rural areas showed higher positive cases than the centers of cities which reached 86 case (69.35%) and the cities centers cases were 38 case (30.65%). Also the infected cases were classified according to the jobs, the results revealed that house wives cases were higher than other jobs reached 5 case (4.9%), while childhood cases were the lowest cases reached 6 cases (4.9%). While classification according to the sex, results revealed that females cases were higher than males cases 63 female cases (50.80%) while male cases were 6(49.0%). Also results classified according to the age, the range from (544 year) had the highest percent 75 case (60.48%) while the range from 65 and more had the lowest cases, cases (.6%). While classification according to the months revealed that june showed the highest cases which reached 0 case (6.%) while December showed the lowest cases case (0.80%). The results of the positive rose bungal tests in sheep during the five years of the study were 349 case, the highest infected cases appeared in 993 which reached 364 case (34.6%) while 997 showed the lowest infected cases which reached 340 case (0.49%). The number of aborted cases which were register during the same period were 707 case (0.4%) and in 993 the results showed the highest percent of abortion 483(0.43%) while 996 showed the lowest number, 40(5.6%). The results revealed that the infected cases of humans and animals were corresponding from the geographical distribution and according to the months, this pointed that the main source for the humans infections were brucella infected animals. We are concluded that brucelloses was endemic in AlHashymia district and it causes large economic loses and health problems. So the disease must be controlled in the animals by using a safe vaccine for all domestic and wild animals. 0
